Infectivity of Maize Chlorotic Mottle Virus from Contaminated Maize Seeds

Seeds have been identified as major sources of introduction and spread pathogens, with viruses being detected in the seed also on coat. In this study, infectivity Maize chlorotic mottle virus (MCMV) through seeds was investigated. that had tested positive for MCMV previously using double antibody enzyme-linked immunosorbent assay (DAS-ELISA) real-time reverse transcription polymerase chain reaction (real time RT-PCR) were obtained from various sources. The soaked phosphate buffer overnight solution used to inoculate maize seedlings. whole ground mixed inoculation seedlings by hand rubbing. Visible symptoms observed less than 2% 547 inoculated soak extract contaminated 28 days after confirmed DAS-ELISA. Use real revealed one used. mean cycle threshold (Ct) values samples showed ranged 28.21 29.40 cycles. means significantly different (P<0.001) other tested, healthy negative controls. When compared MCMV-infected leaf sap, there visible development associated infection, a severity score three Ct low 11.53. results show evidence infection caused present extract. Despite rare occurrence infectivity, presence viable may cause field, leading lethal necrosis disease where cereal potyvirus is present.
